Research on SARS-CoV-2 Targeted Detection Kit

1. SARS-CoV-2 proprotein convertase (PC) activity and inhibition analysis kit
The SARS-CoV-2 spike glycoprotein has a furin/PC cleavage site on the boundary of the S1/S2 subunits, which can be cleaved by furin secreted by host cells and bacteria and/or furin-like PCs.
Unlike SARS-CoV, SARS-CoV-2 entering cells is pre-activated by furin and/or furin-like pc, reducing its dependence on target cell protease entry.
It has been proven that
the lysis and activation of S protein is essential for the SARS-CoV-2
spike-mediated virus-ACE2 binding, cell-cell fusion, and virus entry into human
lung cells.
SARS-CoV-2 preprotein convertase (PC) activity and inhibition analysis kit
Significance: Furin and/or PC-based SARS-CoV-2 S1/S2 lysis increases the entry and replication of SARS-CoV-2 into cells and eventually develops higher COVID-19 pathogenicity. Therefore, the detection of SARS-CoV-2 targeting Furin/PC lytic activity will help determine why the sensitivity and pathogenicity of COVID-19 are different in different individuals.
The screening
of inhibitors will help develop effective COVID-19 treatment drugs by blocking
the SARS-CoV-2 lysis at the S1/S2 boundary.

2. SARS-CoV-2 total IgG/IgM and neutralizing antibody measurement kit
SARS-CoV-2 infection usually induces an antibody response.
The antibody can bind to SARS-CoV-2 spikes, and only a small part has a
neutralizing function. Only neutralizing antibodies can block the effect of
SARS-CoV-2 spikes and inhibit the SARS-CoV-2 virus from infecting new cells.
SARS-CoV-2 total IgG/IgM and neutralizing antibody measurement kit
Significance: Measuring the total SARS-CoV-2 specific IgG/IgM helps determine whether an individual is infected with SARS-Cov-2.
However, it is more important to detect SARS-CoV-2 neutralizing antibodies in a quantitative and high-throughput format:

3. SARS-CoV-2 Spike-ACE2 binding activity and inhibition analysis kit
The process of SARS-CoV-2 entering human host cells occurs through the binding of the surface unit S1 of its spike protein to the cell receptor angiotensin converting enzyme 2 (ACE2).
ACE2 is ubiquitous and is widely expressed at different levels in the lungs, heart, kidneys, blood vessels, brain, intestines and testes.
ACE2 also mainly binds to cell membranes and is usually less present in the circulation. It has been found that the increased expression of ACE2 in the nasal epithelium and lungs is related to the higher rate of SARS-CoV-2 infection and the severity of COVID-19, respectively.
A new observation suggests that increased circulating ACE2 concentrations may be related to increased incidence and mortality of COVID-19.
In addition, SARS-CoV-2 invasion will degrade ACE2 on the cell membrane and may
increase the concentration of soluble ACE2 in the circulation.

Significance: SARS-CoV-2-ACE2 binding activity determination will truly reflect the level of functional ACE2 in binding to SARS-CoV-2.
This assay can not only detect ACE2 binding activity under healthy and diseased conditions, but also measure changes in ACE2 levels under various experimental interventions, including in vivo ACE2 epigenetic regulation, in vivo and in vitro ACE2 activity inhibition (such as: antibodies and Small molecule inhibitors) and environmental impact (e.g.: smoking).
